Cardiff School of Medicine invites applications for a PhD studentship funded by the Arthritis Research Campaign.

Project Title: Death Receptor 3: a Regulator of Bone Turnover and New Target for Therapy for Osteoporosis'

Supervisors: Dr A S Williams (Department of Rheumatology) and Dr ECY Wang (Department of Medical Biochemistry and Immunology), Cardiff School of Medicine

Project Description: Osteoporosis is a disease which affects our bones, it causes them to become brittle and when this happens they are more likely to break. We are not entirely sure how a normal, strong health bone becomes so weak but it is likely caused by an imbalance between two specialist cells found in bone itself. These cells are called osteoclasts (they break down bone) and osteoblasts (which make new bone), normally they work together to help bones grow and remain strong. In osteoporosis the osteoclast takes up the driving seat and steers bone towards destruction whilst the osteoblast switches to ‘snooze’. We do not know what stops the osteoclast from working in tandem with the osteoblast. There may be a communication breakdown. We know that osteoclasts and osteoblasts contact immune cells using chemical messengers (known as cytokines). These cytokines stick to ‘docking’ (receptors) found on the surface of target cells. When the cytokine and its receptor come together, like two pieces of a jigsaw, the target cells respond by dividing, making protein, dying or even changing to a different cell type, for example. There are many cytokines and cytokine receptors in our bodies and new ones are being discovered all the time but which ones are important in bone' We think that the cytokine, TNF-like protein 1A (TL1A) and its receptor, Death Receptor 3 (DR3) trigger crucial effects on bone cells which determine their function and fate.

This project will look at how DR3 and TL1A control osteoclasts and osteoblasts in osteoporosis using clinical samples and well defined experimental models. Whilst the PhD student is conducting his/her research project they will be expected to meet four objectives:

1. To establish the mechanism by which DR3/TL1A turns immune cells into bone resorbing osteoclasts
2. To determine whether the DR3/TL1A axis drives bone towards osteoporosis and osteoporotic fracture
3. To assess the role of DR3/TL1A in osteoblasts
4. To study DR3 dependent changes in bone density in vivo.

Drs Williams and Wang will meet with the student on a weekly basis to identify outcomes through an accurate analysis of research data, formulate hypotheses (using models and other techniques to explore them) and then make a reasoned selection of methods for achieving the quality of outcomes required. Together we will assess the effectiveness of strategies, identifying factors that had an impact on the outcomes.

Funding Amount: The Studentship will provide full tuition fee support at the UK/EU level, plus an annual stipend (£17,334 for 2009/10, with annual increases thereafter) and a £5,000 award towards project consumables per year.
